Preparation and Deglycerolization of Frozen Red Blood cells



Abstract
We tried to analyze the status of 5 units of frozen-thawed-deglycerolized red blood cells for the evaluation of practical applicability. After whole blood and packed red cells were stored for three days in CPDA-1 con
taining blood bag, those were
frozen
in -80¡É refrigerator by high glycerol technique, and thawed at 37¡Éwater bath, and then deglycerolized in the cell washer. In the course of deglycerolization, most of the leukocytes and platelets were removed compared with those of unfrozen
status.
The mean recovery rate of red blood cell was 73% and the values of osmolarity were within normal limits, whereas the level of plasma hemoglobin and potassium were relatively high.
We suggest that it is necessory to increase the washing volume during the cell washing process in order to obtain the appropriate deglycerolized red blood cells. Although the recovery rate of deglycerolized red blood cells was slightly lower than
those
recommended by AABB, the deglycerolized red blood cells may be useful in a critical situation such as wartime.
